Determination for Blood Concentration of Oxcarbazepine Active Metabolite by Ultra-High Performance Liquid Chromatography
Objective: To establish the ultra-high performance liquid chromatography method for the blood concentration determination of active metabolite of oxcarbazepine 10
11-dihydro-10-hydroxyl carbamazepine. Methods: Venous blood samples were collected
proteins were precipitated by perchloric acid
and the supernatant was centrifuged and fed into the sample. The determination was performed on Waters ACQUITY UPLCBEH C18 (2.1 mm×50 mm
1.7 μm) with 0.05% formic acid aqueous solution∶acetonitrile =75∶25 as mobile phase. The detection wavelength was 254 nm
the flow rate was 0.20 mL/min
and the column temperature was 40 ℃. Results: The linearity of the peak area of 10
11-dihydro-10-hydroxyl carbamazepine in the concentration range from 5 to 100 μg/mL and the peak area ratio of the internal standard to the concentration ratio were good (r=0.999
9)
and the recoveries
precision
stability and accuracy were within the limit requirements. Conclusion: The method is simple
accurate and sensitive
which can be used for blood concentration monitoring of active metabolite of oxcarbazepine 10
