Effect of salidroside on the TGF-β1/Smads signaling pathway in rat liver tissue
Objective:To explore the effect of salidroside on the expression of transforming growth factor-β1(TGF-β1)
Smad2/3
p-Smad2/3
and c-ski in rat’s liver injued by hepatic artery ligation(HAL).Methods:Total 66 Wistar rats were randomly divided into three groups:control group
ligation group
and salidroside treatment group. The time of ligation was divided into five section as 1
2
4
6
and 8h.In the salidroside treatment group
rats were intraperitoneally injected with salidroside solution at 2 days
1 day
and 2 hours prior to ligation.ALT
AST
and ALP in serum were detected by automatic biochemistry analyzer.Pathological changes in liver tissue were observed by HE staining. The protein expressions of TGF-β1
Smad2/3
P-Smad2/3
and c-ski were detected by immunohistoche-mical staining
and the mRNA expression of TGF-β1
Smad2
Smad3
and c-ski were detected by RT-PCR.Results:The levels of ALT
AST
and ALP in serum were improved and the pathological changes of liver tissues were obviously perfect in the Salidroside treatment group. The protein and mRNA of TGF-β1
Smad2/3
and the protein of p-Smad2/3 in the ligation group were strongly more positively expressed than those in the control group(P<0.01)
and salidroside could decrease the expression of TGF-β1
Smad2/3
and P-Smad2/3(P<0.05).The protein and mRNA of c-ski in the ligation group were less expressed than those in the control group(P<0.01)
but salidroside could increase the expressions of c-ski(P<0.05).Conclusion:Salidroside has significant effect on the TGF-β1/Smads signaling pathway in rats with hepatic artery ligation. It could inhibit activation of TGF-β1/Smads signaling pathway and protect acute liver injury. The mechanism may be inhibiting the expression of TGF-β1and Smad2/3 and preventing Smad2/3 phosphorylation as well as up-regulating the c-ski expression.
