摘要

目的:观察隐丹参酮（Cryptotanshinone

CTN）对谷氨酸（Glutamate

Glu）损伤体外原代培养大鼠皮层神经细胞的保护作用并探讨可能的机制。方法:体外培养大鼠大脑皮层神经细胞

建立Glu损伤模型

采用MTT法检测细胞存活率

DCFH-DA染色法检测细胞氧自由基水平

Hoechst荧光染色检测细胞凋亡

Western blot检测Bcl-2和Bax的表达。结果:CTN能明显减轻Glu对皮层细胞的损伤

提高细胞的存活率

降低细胞内的活性氧水平

显著上调Bc1-2蛋白表达

减少Bax蛋白表达

抑制细胞凋亡。结论:CTN对Glu损伤神经细胞有显著的保护作用

其机制可能与抑制或清除Glu诱导的细胞内活性氧的生成

调节凋亡相关基因Bcl-2蛋白家族的表达有关。

Abstract

Objective:To observe the protective effect of cryptotanshinone on glutamate-induced apoptosis in cultured cortical neurons.Methods:Cell viability was detected by MTT assay.Intracellular reactive oxygen species(ROS) level was detected using DCFH-DA.The apoptosis rate was measured by Hoechst 33342

and the expression of Bcl-2 was detected by Western blot.Results:Compared with glutamate group

cryptotanshinone could increase cell survival rate

decrease apoptosis rate and the level of intracellular ROS.Moreover

cryptotanshinone could remarkably increase the expression of Bcl-2 protein and decrease Bax protein.Conclusion:Cryptotanshinone could significantly protect cortical neurons from the apoptosis induced by glutamate

the mechanisms of which may be the inhibition or decrease of ROS formation

and increase of Bcl-2 expression.
