摘要

目的:探讨cAMP反应元件结合蛋白（Cyclic adenosine monophosphate response element binding protein

CREB）基因多态性与抑郁症及自杀行为的相关性。方法:运用聚合酶链反应（Polymerase chain reaction

PCR）及限制性片段长度多态性（Re-striction fragment length polymorphism

RFLP）的方法检测50例抑郁症患者（患者组）和50名健康对照者（对照组）的CREB基因上单核苷酸多态性（Single nucleotide polymorphism

SNP）rs10932201、rs6740584-、835 C→A的等位基因和基因型分布情况

确定其与抑郁症及自杀行为是否相关。结果:3个SNP位点符合Hardy-Weinberg遗传平衡度检验。rs10932201和rs6740584位点的基因型和等位基因在抑郁患者和正常对照组中差异无统计学意义。-835 C→A位点在病人组和对照组之间差异无统计学意义;但将病人分层后

其C/A基因型在有自杀行为的抑郁患者组和对照组中差异有统计学意义（χ2=5.1409

P=0.023 4）。结论:汉族人群中CREB基因启动子区域-835C→A多态性可能与有自杀行为的抑郁症存在相关性。

Abstract

Objective:To explore the association between polymorphism of the cAMP-responsive element binding protein(CREB)gene promoter region

major depressive disorder and suicidal behaviors.Methods:The allele and genotype distribution of CREB gene SNP rs10932201

rs6740584

-835 C→A were detected in 50 depression patients and 50 healthy control subjects by using polymerase chain reaction(PCR)and restriction fragment length polymorphism(RFLP)

so as to determine the association between CREB and depression.Results:The gene polymorphism at rs10932201

rs6740584

and-835 C→A were indefinite.However

the significant difference in C/A genotype distribution at-835 C→A was found between the depressive patients with suicidal behavior and control group(χ2=5.140 9

P= 0.023 4).Conclusion:The findings support that the gene polymorphism of-835C→A at CREB gene promoter region in Han nationality is associated with depressive patients who have suicidal behavior.
