Mechanisms of TMP inhibiting cell migration in SKOV3 cells induced by extraneous IL-8
Objective:To investigate whether tetramethylpyrazine(TMP)has the activity of inhibition on cell migration induced by extraneous Interleukin-8(IL-8) in SKOV3 cell
and to understand its probable mechanisms. Methods:MTT was used to determine the concentrations of TMP.The wound healing assay and transwell assay were used to study the cell migration. The protein expression levels of E-cadherin and matrix metalloproteinase 9(MMP-9) were detected by Western blot.Results:IL-8(100 ng/ml)increased the ability of cell migration in SKOV3 cell
TMP(50
100 μg/ml)inhibited this process.IL-8 down-regulated the protein expression of E-cadherin(40.2±5.3)%
and up-regulated the protein expression of MMP-9(34.6±4.4)% respectively and significantly
and TMP(50
100 μg/ml)increased the expression of E-cadherin(74.9±6.4)% and decreased the expression of MMP-9(86.5±5.8)% in SKOV3 cells. Conclusion:TMP inhibits cell migration induced by extraneous IL-8
which correlates with variable protein expression changes of E-cadherin and MMP-9 in SKOV3 cells.
