Investigation into the effect and mechanism of Hedgehog signaling pathway on invasion and migration of human gastric cancer cell line SGC-7901
Objective:To investigate the effect and mechanism of Hedgehog signaling pathway on invasion and migration of human gastric cancer cell line SGC-7901.Methods: After cyclopamine blocked Hh signaling pathway specifically
transwell chamber assay and wound healing assay were used to examine the capacities of invasion and migration of SGC-7901 cells respectively.Reverse transcriptase-polymerase chain reaction(RT-PCR) was used to detect the expression of MMP-2 and MMP-9 mRNA in SGC-7901 cells.Immunochemistry was used to detect the expression of MMP-2 and MMP-9 protein in SGC-7901 cells. Results: The capacities of invasion and migration of SGC-7901 cells could be significantly inhibited after blocking Hh signaling pathway(P<0.05).There was lower expression of MMP-2 and MMP-9 mRNA than the untreated group in SGC-7901 cells(P<0.05).The expression of MMP-2 and MMP-9 protein also remarkably decreased(P<0.05).Conclusion: Hedgehog signaling pathway may promote invasion and migration of SGC-7901 cells by regulating the expression of MMP-2 and MMP-9.
