Effect of electrical stimulation on the expressions of LINGO1 and NgR1 in the brain of rats after focal cerebral infarction
Abstract
Objective:To explore the mechanism of electrical stimulation improves motor recovery in a rat stroke model.Methods:Spragne-dawley(SD)rats were subjected to establish the stroke model and randomly divieded into ischemic group
electrical stimulation group
sham operation group and normal group.Electrical stimulation was used 90 min after the operation
30 min/d.The expressions of LINGO1 and NgR1 were measured with RT-PCR and immunohistochemistry at 1 d
3 d
7 d
14 d
21 d respectively after the ischemia. Results:Motor function significantly recovered in electrical stimulation group compared with the ischemic group(P<0.05).The expressions of LINGO1 mRNA and protein in ischemic group reached the peak at 1d and were still higher than sham operation group at 21 d(P<0.01).The expression of LINGO1 protein in electrical stimulation group was lower than ischemic group at 7 d(P<0.05) but the expressions of LINGO1 mRNA and protein were higher in electrical stimulation group than in ischemic group at 21d(P<0.01).The expressions of NgR1 mRNA and protein in ischemic group elevated at 1d(P<0.01)
reached the peak at 3 d(P<0.01) and returned to normal at 14 d(P>0.05).The expressions of NgR1 mRNA and protein in electrical stimulation group were lower than ischemic group at 3d and 7 d(P<0.05).Conclusions:The expressions of LINGO1 and NgR1 in the ischemic cortex increased significantly. The decreased expressions of LINGO1 protein
NgR1 mRNA and NgR1 protein induced by electrical stimulation might be one of the mechanisms of motor recovery after acute cerebral infarction.
