Comparative proteomics analysis of cytoplasmic proteins of immortalized cervical cell and cervical cancer cell
Objective:To identify differentially expressed protein spots of cytoplasm from H8 cells(human immortalized endocervical cell) and Caski(cervical cancer cell) associated with cervical carcinogenesis by two-dimensional electrophoresis(2-DE) analysis. Methods:The cytoplasmic proteins of H8 cell and Caski cell were separated by means of two-dimensional gel electrophoresis
then the selected differential protein spots were analyzed by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF-MS) and SWISS-PROT database.Results:6 differentially expressed protein spots were successfully identified
among which
S100P
IKKb(Inhibitor of nuclear factor kappa-B kinase subunit beta)
TRAF5(TNF receptor-associated factor 5) were significantly up-regulated in cytoplasmic proteins in Caski cell.STK3(Serine/threonine-protein kinase 3) deletion was found in cytoplasmic proteins in Caski cell. Conclusion:The differential expressions of proteins of cytoplasm may be valuable for studying mechanisms of cervical carcinogenesis
or as diagnostic markers and therapeutic targets for cervical cancer.
