The influence of slidroside to wnt signaling pathway in rat hepatic stellate cells stimulated by acetaldehyde
Objective:To explore the role of wnt signaling pathway and the effect of salidroside on wnt pathway and cell proliferation in rat hepatic stellate cells
we detected the diversity of mRNA
protein expression of Wnt signaling pathway downstream molecules and the proliferation of rat hepatic stellate cells treated by acetaldehyde and salidroside. Methods:We treated the rat hepatic stellate cell with salidroside and acetaldehyde
drawn the cell growth curve using the method MTT
β-catenin
Collagenα1 and Cyclin D1 were detected by reverse transcription PCR and Western blot. Results:By comparing the cell growth curve
we found clearly that proliferation rate of HSCs stimulated by acetaldehyde can be inhibited after salidroside treatment. We also found that mRNA and protein synthesis of Collagenα1
Cyclin D1 and β-catenin were enhanced in rat hepatic stellate cell stimulated by acetaldehyde[1]
however the mRNA expression and protein synthesis of Cyclin D1
β-catenin were declined in the cells treated by salidroside than cells stimulated by acetaldehyde only. Conclusion:The classical wnt signaling pathway may be involved in activation of hepatic stellate cells stimulated by acetaldehyde. The salidroside may reduce the proliferation of rat hepatic stellate cell stimulated by acetaldehyde through inhibiting the wnt signaling pathway.
