Effect of GRP78 on invasion and metastasis of human gastric carcinoma SGC7901 cells in vitro
Objective:To investigate the effect of BAPTA-AM and A23187 on GRP78 expression of gastric carcinoma SGC-7901 cells in vitro
and the effect of GRP78 on invasion and metastasis of these tumor cells. Methods:The SGC7901 cells were treated by BAPTA-AM and A23187.RT-PCR and Western blot were used to assess the expression of GRP78 at both mRNA and protein levels.Matrigel invasion assay and a scratch wound assay were used to assess the invasion and migration levels. Results:The expression of GRP78 at both mRNA and protein levels
the invasion and migration levels were decreased in the BAPTA-AM-treated cells and increased in A23187-treated cells as compared with those of control group.Conclusion:BAPTA-AM can down-regulate the expression of GRP78 in SGC7901 cells and A23187 can up-regulate it.The expression of GRP78 in SGC7901 cells may be associated with their migration and invasion levels.
