Effects of salvianic acid A(danshensu) on proliferation, activation and TGFβ/BMP receptors in hepatic stellate cells
Objective:To investigate the effects of salvianic acid A(Danshensu) on proliferation
activation
expressions of TGFβand BMP receptors in hepatic stellate cells
and the possible mechanism.Methods:LX-2 cells were cultured in vitro
and devided into 5 groups:normal control as group A
5 ng/ml TGFβ1 treatment as group B
and salvianic acid a groups with 0.25
0.125
0.0625 mmol/L of salvianic acid A and 5 ng/ml TGFβ1 as groups C1-3.After 48h treatment
cell proliferation was measured by Cell Counting Kit-8.The expressions of α-smooth muscle actin(α-SMA) was detected by the immunocytochemical assay.Expressions of TGFβreceptor Ⅰ
Ⅱ(TβRⅠ
Ⅱ) and BMP receptorⅡ(BMPRⅡ) mRNAs
were detected by RT-PCR.Results:The proliferation of LX-2 treated by TGF-β1 was not significant different from the control group
salvianic acid A suppressed the proliferation of TGF-β1 treating LX-2 in a dose-dependent manner(P<0.01)
and also the TGF-β1 inducing activation.Expressions of TGFβreceptors decreased and expressions of BMP receptors increased dose-dependently(P<0.01) after treated by salvianic acid A for 48 h.Conclusion:salvianic acid A inhibits the proliferation and activation of HSCs possibly by down-regulating the TβRⅠ
Ⅱ expressions and antagonizing TGFβinduced down-regulating the BMPRⅡ expressions.
