摘要

目的:探讨不同的光密度（Optical density

OD）值、离心速度及时间对大肠杆菌透射电镜样品制备及超微结构观察的影响。方法:按照正交试验设计原则

将不同OD600值的大肠杆菌分成9个不同的离心速度和时间组

比较各组制样难度及电镜下超微形态及纵、横切面比的变化。结果:各种OD600值和离心条件对大肠杆菌超微结构影响不大

但OD600值为0.8

离心时间为15min时

样品中细菌多呈纵切形态而更适宜观察分析。结论:离体培养大肠杆菌透射电镜样品制备及观察的最佳OD600值为0.8

离心时间为15min

离心速度为5000～12000r/min。

Abstract

Objective:To investigate the influences of different OD value and centrifugal parameters on the ultrastructure of Escherichia coli in order to find out the best OD value

speed and time of centrifugation.Methods:According to orthogonal experimental designing principle

the E.coli with different OD600 values are divided into 9 different groups of centrifuge speed and time.The changes of E.coli morphology in each group at different OD600 value

centrifuge time and speed were compared under transmission electron microscope.Results:The different OD600 values and centrifugal parameters have little influence on the ultrastructure of E.coli

but when the OD600 value at 0.8

centrifuge time at 15 min

the cross-section ratio was significantly higher than other groups and was more suitable for observation.Conclusion:The best parameter to prepare the TEM samples of E.coli is under the OD600 value at 0.8 and centrifugation time at 8 minutes and speed at 5 000～12 000 r/min.
