摘要

目的:检测抗凋亡基因survivin在肾透明细胞癌786-O细胞株中的表达并探讨其临床意义。方法:运用MTT（四唑盐比色）法检测细胞生长状态并绘制生长曲线

运用RT-PCR（逆转录聚合酶链反应）法和免疫组化法检测786-O细胞株中survivin mRNA及survivin蛋白的表达情况。结果:检测出786-O细胞株中有survivin mRNA的表达

survivin蛋白的表达亦呈阳性

而正常肾细胞无阳性表达。结论:survivin mRNA及survivin蛋白在肾透明细胞癌中均有明显的表达

利用这一表达特异性

为以survivin基因为靶点的肾癌基因治疗研究提供了实验依据。

Abstract

Objective:To investigate the expression and the clinical significance of survivin in renal clear cell carcinoma 786-O.Methods:The expression of survivin mRNA and protein were detected by RT-PCR and immunohistochemisty

The growth curve of renal clear cell carcinoma 786-O were detected by MTT.Results:The survivin mRNA and protein were detected in 786-O cells.In contrast

no expression of survivin was found in normal renal cells.Conclusion:The survivin gene and protein were significantly expressed in renal clear cell carcinoma 786-O.This may provide the experimental evidence for the survivin gene as a target of gene therapy in renal cancer.
