摘要

目的:分离纯化茅莓全草中的一种皂苷类成分—悬钩子皂苷R1。方法:采用大孔吸附树脂与制备型反相高效液相色谱组合分离技术

将茅莓水提醇沉后的粗制品经大孔吸附树脂柱色谱预分离后

再用100～200目硅胶柱色谱分离

用氯仿-甲醇-水-冰醋酸（40:10:1:0.4%）洗脱

收集样品进行制备型RP-HPLC纯化精制。结果:产品经熔点、MS、IR、NMR、UV和HPLC图谱鉴定

并与文献比较

证明为悬钩子皂苷R1

其纯度>98%。结论:该方法较为简单可行

重复性好

采用的溶剂系统价廉易得、沸点低、易于回收;悬钩子皂苷R1产品可用做分析方法的对照品。

Abstract

Objective:To develop a method for the isolation and purification of suavissimoside R1 from Herb Rubi Parvifolii. Methods:Macroporous adsorption resin and preparative RP-HPLC

the water extract sedimentated by ethanol was pre-isolated by adsorption resin

and then fractionated by column chromatography on silica gel ( 100-200 mesh )using chloroform-methanol-water-acetic acid(40:10:1:0.4%).The collected fraction was purified for suavissimoside R1 by preparative RP-HPLC were combined as pre- pareation method.Results:Suavissimoside R1 was identified by mass spectrometry(MS)

infrared spectrometry(IR)

ultraviolet spectrometry(UV)

nuclear magnetic resonance (NMR) and high performance liquid chromatography(HPLC)

and compared with the relevant literature.The final purity of the compound was more than 98%.Conclusion:The developed method is simple

and reproducible.The solvent is cheap

low in boiling point and easily recovered.The product of suavissimoside R1 can be used as reference substance for analysis.
