Effect of polycystic ovary-like environment on E2 and P levels in human granulosa cells in vitro
Objective:To study the effect of polycystic ovary-like environment on secretion of E
2
and P from human granulosa luteal cells in vitro. Methods:According to different additives granulosa cells cultured in vitro which were retrieved from infertility women with normal ovulation who were undergoing in vitro fertilization-embryo transfer(IVF-ET)
were divided into four groups:A (androstenedione) group
LH group
INS (insulin) group and A+LH+INS group. Each group was subdivided into polycystic ovary-like environment group with higher levels of the additives and physiological environment group with lower levels of the additives.Levels of E
2
and P in medium were detected separately. Results:Levels of E
2
and P in polycystic ovary-like environment were significantly decreased in A group and A+LH+INS group
and significantly increased in LH group and INS group (P
<
0.01 respectively).Levels of E
2
and P of LH group
INS group and A+LH+INS group were all significantly higher than that of A group
with the highest level of E
2
in INS group and the highest level of P in LH group. Moreover
compared with INS group or LH group respectively
concentrations of E
2
and P in A+LH+INS group were both significantly decreased in polycystic ovary-like environment but no difference was found in physiological environment. Conclusion:In polycystic ovary-like environment
E
2
and P secretion from normal granulosa luteal cells may be inhibited by A.
