Growth-inhibiting effects of oxaliplatin on K562 cells in vitro
Objective:To investigate the effects of oxaliplatin on human leukemia K562 cell and its mechanism. Methods:The leukemia cell growth was measured by MTT-microculture tetrazolium assay.Cell-cycle kinetics and apoptosis were analyzed by flow cytometry and microscopic examination. Results:Oxaliplatin inhibited the leukemia cell growth in concentration- and time-dependent manners.Marked cells accumulated in G
2
/M phase. The rate of cell growth inhibition was 85% after intervention in 0.016 mmol/L oxaliplatin for 72 hours. The cells showed G
2
/M arrest and apoptosis. Conclusion:Oxaliplatin suppresses the growth of K562 cells in vitro by causing cell-cycle arrest
and cell apoptosis. The mechanisms of oxaliplatin provide theoretical guidance for clinical treatment.
