Establishment and MDR characteristics of HepG2/Adm nude mice xenograft
Objective:To establish an animal model of HepG2/Adm nude mice xenograft and investigate the mechanism of multidrug resistance(MDR).Methods:HepG2/Adm cells were transplanted into the subcutaneous tissue in athymic nude mice. The differences in the MDR characteristics between HepG2/Adm and nude mice xenografts(HepG2/Adm/nude)were investigated.Cytotoxicity was determined with MTT assay. The expression of MDR-related gene protein and mRNA was examined by flow cytometry(FCM)and reverse transcriptase polymerase chain reaction (RT-PCR) respectively. Results:Tumor incidence was 100% after subcutaneous implantation with 10
9
of cells.IC
50
values (the concentration producing 50% maximal inhibition) of Adriamycin(ADM) to HepG2/Adm and HepG2/Adm/nude were 4.21 μg/ml
4.10μg/ml
respectively(P
>
0.05).The expressions of MDR protein (P-gp
MRP and LRP)in HepG2/Adm and HepG2/Adm/nude were 92% and 91.8%
88.7% and 88.1%
81.1% and 80.7%
respectively(P
>
0.05).The expression of MDR-related gene mRNA in HepG2/Adm/nude had no significant differences from that of HepG2/Adm(MDR) (P
>
0.05).Conclusion:HepG2/Adm/nude xenograft established by HepG2/Adm cells transplantation retains the characteristics of MDR phenotype. It can provide an ideal animal model for the study of human tumor MDR.
