Development of ischemic preconditioning model of cultured neonatal rat cardiomyocyte in vitro
Objective:To make a convenient and practical ischemic preconditioning model in cultured neonatal rat ventricular myocytes. Methods:The cultured cardiomyocytes dissociated from neonatal rat ventricle were placed in a modified chamber and mixture gas(95%N 2
5%CO 2)or 100% N 2 was put inside. The injury of cardiomyocytes was assessed by LDH release and trypan blue exclusion in four groups(n=6 in each)—Group control-1
Group IPC-1
Group control-2 and Group IPC-2.Results:The release of LDH was lower and cell viability was higher in Group IPC-1 than Group control-1.Similar results were obtained in Group IPC-2 compared with Group control-2.The difference in the release of LDH and cell viability was not significant between Group IPC-1 and Group IPC-2 (P>0.05).Conclusion:The isehemic preconditioning model in vitro may be induced easily by oxygen deprivation in cultured neonatal cardiomyocytes
and can be used to further studyon the mechanism of the late phase of preconditioning.
