摘要

目的 :探讨大鼠心肌细胞缺血预处理时COX2 的作用及其与P38MAPK的关系。方法 :实验分为 4组（对照组、缺血预处理组、SB组和NS3 98组）

用Westernblotting法测定心肌细胞的COX2 表达量和Phosph -P38MAPK量

以LDH释放和台盼蓝排斥试验判断心肌细胞损伤程度。结果 :缺血预处理组的LDH释放较对照组少、细胞存活率则较高 ;SB组和NS3 98组LDH释放较缺血预处理组多、细胞存活率则较低。NS3 98组的Phosph -P38MAPK量与缺血预处理组无差异（P >0 .0 5 ）

SB组的COX2 表达量比缺血预处理组低

差异显著（P <0 .0 1）。结论 :缺血预处理时COX2 和P38MAPK表达、活化

COX2 活化依赖P38MAPK

在其信号传导中

P38MAPK活化是COX2 的上游。

Abstract

Objective:To explore the association of COX 2 with P38MAPK in late phase of ischemic preconditioning.Methods:The expression of COX 2 and P38MAPK activity were determined by Western blot technique and the injury of cardiomyocytes was assessed by LDH release and trypan blue uptake in four groups( n =4 in each)-Group control

Group IPC

Group SB and Group NS 398 .Results:The signal of COX 2 and amount of phosph-P38MAPK were higher in Group IPC than those in Group control.The expression of COX 2 in Group IPC increased markedly in comparision with Group SB

however the difference in the amount of phosph-P38MAPK was not significant between Group NS 398 and Group IPC ( P >0.05).Conclusion:In late phase of ischemic preconditioning

the P38MAPK activity and COX 2 expression increase significantly in rat cardiomyocyte

P38MAPK mediated COX 2 expression

and may be an up-stream of COX 2.
