NF-κ B inhibitor-PDTC enhance TNF-α induced cytotoxicity in breast cancer cells
Objective:To evaluate the effect of NF-κ B inhibitor-PDTC on TNF-α induced cytotoxicity in breast cancer cells. Methods:The cell survivals of breast cancer cell lines MCF-7 and MDA-MB-435s were measured by MTT assay in vitro.MDA-MB-435s cells were implanted into BALB/C mice. Animal tumor models were treated with TNF-α
PDTC and TNF-α plus PDTC.Results:The growth of two breast cancer cell lines was not inhibited by either TNF-α(2000U/mi or less)or PDTC.However
both breast cancer cells treated with TNF-α(2000U/mi)plus PDTC(50μ mol)showed significant growth inhibition as compared with controls. In vivo
the growth of implanted tumor was significantly reduced in volume in mice treated with TNF-α(7.5×10 6U/kg
ip
bid) plus PDTC(50mg/kg
ip
bid) as compared with controls( P <0.01).Tumor growth was not inhibited by either TNF-α or PDTC treatment.Conclusion:NF-κ B inhibitor-PDTC evidently enhance TNF-α in inducing cytotoxicity in breast cancer cells
and thus inhibit the growth of cells.
