Objective :This study is to transplant hepatocyte subcutaneosly and to see the possibility of eliminating CMXR by using this new model. Methods Preparation of isolated hepatocytes was performed fundamentally according to the collagenase digestive method of Berry and Friend using a recirculating perfusion apparatus. Hepatocellular transplantation was carried out by direct injection of the hepatocyte suspension into the splenie parenchyma or injecting subcutaneously. Recipients were killed
and their spleen and skin specimens were fixed in 10% formalin and stained with hematoxylin and eosin and for PAS. For G - 6 - P and SDH stain
fresh thin frozen sections were used. Results The results revealed that no significant difference of the xenograft survival was found between splenic hepatocyte and subcutaneous hepatocyte within one month. It is always believed that the CMXR happen from four to seven days after transplantation. Conclusion So the subcutaneous hepatocellular transplantation can be used as the model to appreciate CMXR.
