摘要

目的观察三苯氧胺（Ｔａｍｏｘｉｆｅｎ，　ＴＡＭ）及雌激素（Ｅ２）对乳腺癌ＭＣＦ－７细胞株耐药基因ｍｄｒ－１ｍＲＮＡ水平的影响；方法应用ＲＴ－ＰＣＲ法观察ＴＡＭ及Ｅ２在不同时间、浓度作用下细胞内ｍｄｒ－１ｍＲＮＡ水平的变化；结果ＴＡＭ可以使ＭＣＦ－７细胞内ｍｄｒ－１ｍＲＮＡ水平降低，而Ｅ２则相反。且以上变化表现出剂量、时间依赖关系。结论ＴＡＭ通过降低ｍｄｒ－１ｍＲＮＡ水平，使肿瘤细胞的耐药性降低，有利于抗肿瘤药物更好的发挥作用。

Abstract

Objective Study the effects of Tamoxifen(TAM) and estrogen(E2) on the mdr-1 mRNA level in MCF -7 cell. Methods RT-PCR was used to observe the variation of mdr-1 mANA level in MCF-7 cell after treatment with TAM and E2 at different time and concentration. Results TAM could decrease the level of mdr-1 mRNA in MCF-7 cell but E2 could not. These results came together with the relationship with the concentration of the drug and the treatment of the given time. Conclusion The drug-resistance of neoplasm cells could be down -regulared by TAM via decreasing the mRNA level in cells

which would be helpful in tumor drug therapy.
