Effect of Tanshinone on distribution of Levofloxacin in uterine tissues of pelvic inflammatory rat models
Objective:To study the effect of tanshinone on distribution of levofloxacin in uterine tissues of pelvic inflammatory rats model. Methods:Totally 96 rats in pelvic inflammatory model were randomly divided into experimental group and control group with48 rats in each. Experimental group received tanshinone(8 mg/kg)while rats in the control group were administered with the same volume of saline. After 1 h
rats in both groups were given levofloxacin orally(80 mg/kg). Six rats in both groups were sacrificed for collecting plasma and uterine tissues respectively at the time points of 0.25

8.0 and 12.0 hour post the administration of levofloxacin. Concentrations of levofloxacin in plasma and uterine tissues were determined by high performance liquid chromatography(HPLC) and the pharmacokinetics parameters were calculated with DAS2 software program. Results:The concentration curves based on drug-time in experimental and control group both fitted two-compartment model. The main pharmacokinetics parameters in plasma and uterine tissues were listed as follows:Cmaxwere(14.341±1.551)μg/m L and(12.780±1.062)μg/m L for plasma
(13.910±1.704)μg/m L and(11.671±1.134)μg/m L(P<0.05)for uterine tissues. tmaxwere(1.329±0.269)h and(1.249±0.264)h for plasma
(1.419±0.380)h and(1.280±0.270)h for uterine tissues. t1/2βwere(6.284±1.132)h and(5.442±2.590)h for plasma
(7.920 ±2.802) h and(3.894 ±1.595) h(P <0.05) for uterine tissues. AUC0 →twere(62.667 ±4.516)μg·h/m L and(55.693±3.138)μg·h/m L(P<0.05)for plasma
(55.518±11.709)μg·h/m L and(40.374±2.892)μg·h/m L(P<0.05)for uterine tissues. Conclusion:Tanshinone can increase the distribution of Levofloxacin in the uterine tissues of pelvic inflammatory rat models.
