Primary research of biodegradable PLGA film used for intrauterine adhesion prevention
Objective:Toprimarily research about feasibility of biodegradable poly(lactic-co-glycolic acid)(PLGA)film used for intrauterine adhesion prevention. Methods:PLGA film of different proportions of LA:GA was prepared. The film was immersed into the simulated uterine fluid to test the in vitro degradation. They were also placed in animal uterine lumen to test the in vivo degradation.Thirty Kunming mice were divided randomly into three groups:sham operation
intrauterine adhesion and PLGA film. Masson’s stain and Western blot was used to exam the fibrotic effect of PLGA film on endometrium of uterus. Results:In vitro degradation
the degradation time of PLGA film(50∶50)was the fastest(98.5% weight loss at four weeks)while PLGA film(90∶10)was the slowest(7.8%weight loss at 12 weeks). The degradation time of PLGA film(75∶25)was between PLGA film(50∶50)and PLGA film(90∶10). Most weight of PLGA film(75∶25)was degraded at 4 weeks(95.7% weight loss at 4 weeks). In vivo degradation
PLGA film(50∶50)was degraded completely in 7 days while PLGA film(75∶25)and PLGA film(90∶10)were detained in the uterine cavity after 7 days. PLGA film(75∶25)increased the fibrotic area(P=0.000)and expression of fibrotic proteins(P<0.01)compared with sham operation group in Kunming mice. There was no significant difference in fibrotic area(P>0.05)and expression of fibrotic proteins(P>0.05)in endomtrium between intrauterine adhesion group and PLGAfilm group in Kunming mice. Conclusion :PLGA has a potential application value for intrauterine adhesion prevention
of which degradation time can be regulated without changing the fibrotic degree of endometriumin intrauterine adhesion.
