Effect of inhibition of potassium ions on the proliferation and migration of U-251 cells
Objective:To investigate the effect of inhibition of potassium ions on the proliferation and migration of U-251 cells.Methods:U-251 cells were treated by concentrations of Glib in 12.5 μmol/L to 1 600 μmol/L;the rates of inhibition cells proliferation and screen of concentration were measured by CCK-8 kit;the changes of cells of experimental groups(cells treatment with Glib by100 μmol/L
300 μmol/L and 500 μmol/L)and control group(without Glib)migration and invasion were monitored by wound healing assay and Transwell method. Results:CCK8 assay showed the inhibition of U-251 cell proliferation by Glib range from 100 μmol/L to800 μmol/L and inhibition of U-251 cell proliferation by Glib in a dose-dependent manner
and the median inhibition concentration(IC50)of 48 h was 400 μmol/L(F=150.30
P=0.000). Wound healing assay revealed that the width of experimental groups were bigger than that of the control group at 24 h and 48 h and mobility width rates were negatively correlated with drug concentration(F=130.60
P=0.000). Transwell results showed that the drug inhibition of cell migration ability was consistent with the wound healing results in that the number of migrating cells in the experimental group was significantly fewer than those of the control group(F=86.68
P=0.000).Moreover
Transwell method showed that invasion ability of treatment with glib decreased compared with the control group(F=55.30
P=0.000)
suggesting that the inhibition rates of migration and invasion were dose-dependent. Conclusion:Glib plays an inhibiting role for the proliferation and migration of U-251 cells.
