Effects of curcumin on the expression of TNF-α,NF-κB,COX-2 in rats with autoimmune prostatitis
Objective:To investigate the influence of curcumin on the expression of tumor necrosis factor-α(TNF-α)
nuclear factor κB(NF-κB)(p65)and cyclooxygenase-2(COX-2)in in rats with autoimmune prostatitis. Methods:Sixty male Sprague Dawley rats were equally and randomly divided into four groups:normal control group
model group
curcumin intraperitoneal injection group and celecoxib group(positive control group). The experimental autoimmune prostatitis rat model was made by intraperitoneal injection of vaccine and multiple hypodermic injection of prostate extract mixed with adjuvants. After the experimental animal model being estab-lished
each group was treated for four weeks. The levels of TNF-α in prostate tissues were detected by ELISA assay. The expressions of NF-κB(p65)and COX-2 in rat prostate tissues were respectively measured by immunohistochemistry
immunofluorescence and Western blot. Results:Levels of TNF-α(F=382.73
P=0.000)
NF-κB(p65)(F=56.23
P=0.000)and COX-2(F=122.29
P=0.000)in prostate tissues were significantly higher in model group than in the other groups. Expressions of TNF-a and COX-2 were obviously lower in curcumin intraperitoneal injection group than in positive control group(P<0.01);no significant difference in the expression of NF-κB(p65) was observed between the two groups(P =0.059). Conclusion:Curcumin is effective in treating rats with autoimmune prostatitis
and the mechanism may be associated with the reduction of the cytokines TNF-α
