摘要

目的:定量分析乙肝病毒（hepatitis B virus

HBV）感染的肝癌患者肝内共价闭合环状DNA（covalently closed circular DNA

cccDNA）、DNA和血清中DNA的表达水平

并分析其与乙肝病毒e抗原（hepatitis B e antigen

HBeAg）状态的相互关系。方法:运用特异性荧光定量探针法检测了26例HBV感染的肝癌患者中HBV cccDNA和HBV DNA的表达水平

进一步分析这两种病毒因子在HBeAg阳性和HBeAg阴性样本中的表达差别以及与HBeAg状态的关系。结果:HBV cccDNA和HBV DNA在HBeAg阴性组的表达水平均显著低于HBeAg阳性组（P值分别为0.044和0.0214）

同时

在HBeAg阴性组中

cccDNA的表达水平与DNA是显著正相关的（r=0.918

P=0.000）;HBeAg阳性组中cccDNA与DNA是较弱的正相关关系

且不具有显著性（r=0.591

P=0.072）。结论:肝内cccDNA与DNA表达水平与HBeAg的状态是密切相关的

HBeAg阴性的HCC患者中病毒复制水平下降的原因主要是cccDNA水平的下降。

Abstract

Objective:To quantify intrahepatic cccDNA

DNA

serum HBV DNA and to analyze its relationship with HBeAg in hepatitis B virus-related hepatocellular carcinoma. Methods:The intrahepatic HBV cccDNA and DNA levels in 26 HBV-related HCC patients were measured by real-time PCR

and the differences were compared between HBeAg-positive and HBeAg-negative patient samples. Finally

their relationship with HBeAg status was also analyzed. Results:Intrahepatic HBV cccDNA and DNA levels in HBeAg-negative patients were significantly lower than those in HBeAg-positive patients(P=0.044

P=0.0214). Meanwhile

intrahepatic HBV cccDNA was significantly positively correlated with DNA in HBeAg-negative patients(r=0.918

P=0.000). In HBeAg-positive patients

the positive correlation between cccDNA and DNA was weak and insignificant(r=0.591

P=0.072). Conclusion:Expression levels of intrahepatic cccDNA and DNA is closely related with HBeAg status

and low viremia in HBeAg-negative HCC patients is mainly due to lower cccDNA levels.
