Establishment and evaluation of acute paw edema inflammation model of C57BL/6J mice
Objective:To establish and evaluate acute paw edema inflammation model of C57BL/6J mice. Methods:Twenty-four cases of C57BL/6J male mice were randomly divided into 3 groups. The normal group did not receive any treatment;the other two groups were induced to acute inflammation model by carrageenan. The thickness of the paw was measured respec-tively at 1

5
6 hours after the injection of carrageenan. Then the mice were executed
the serum samples were taken for the detection of inflammatory cytokines
and the foot tissue was taken for pathology detection. Results:At 1 h after the inflammation
the paws of mice in model group began to swell. The edema became significant at 2 h after the inflammation. At 5 h after the inflammation
the edema reached the peak. The edema was significantly inhibited in indomathacin group at 2 h after the inflammation
and the inhibition continued till 6 h after the inflammation. The mice in normal group had no significant change. Compared with those in indomathacin group and normal group
the serum TNF-α and IL-1β of mice in normal group were significantly elevated. The pathological sections of paws of mice in model group showed massive infiltration of neutrophils. Little infiltration of neutrophils was seen in mice in indomathacin group. The paw pathology of mice in normal group showed no obvious abnormality. Conclusion:The acute paw edema and inflammation model of C57BL/6J mice is reliable and highly repetitive. The positive controlling drug indomathacin has obvious anti-edema effect. The model is applicable to the routine screening and evaluation of anti-inflammatory drugs. The changes in paw edema
the increase in serum cytokines TNF-α and IL-1β
and the pathological changes in sites of inflammation are very important indicators to evaluate the acute inflammation model.
