Study on the influencing mechanism of pachymoic acid on myocardial ischemia-reperfusion injury in rats
To investigate the impact of pachymoic acid on myocardial ischemia-reperfusion injury in rats by regulating Shh/Gli1 signaling pathway.
Methods
2
The SD rats were divided into the sham operation group, model group, low dose (5 mg/kg) pachymoic acid group, medium dose (10 mg/kg) pachymoic acid group, high dose (20 mg/kg) pachymoic acid group, and high dose (20 mg/kg) pachymoic acid+cyclopamine (10 mg/kg) group, and the myocardial ischemia-reperfusion model of rats was established. The myocardial infarction was determined by TTC staining, the pathological changes of myocardial tissue in rats was observed by HE staining, the serum levels of aspartate aminotransferase (AST),creatine kinase (CK),and cardiac troponin T (cTnT) were detected by ELISA,the cardiomyocyte apoptosis was detected by TUNEL staining, the expression of Shh and Gli1 of myocardial tissue were detected by RT-PCR; and the expression of Bax, Bcl-2, and Shh/Gli1 signaling pathway related proteins in the rat myocardial tissue were detected by Western blot.
Results
2
Compared with the sham operation group, there were a large area of white infarction area, cardiomyocytes contraction, irregular morphological structure and muscle fiber disorder of the rat myocardial tissue in the model group, with great inflammatory cell infiltration, a increased apoptosis rate, significantly high levels of serum AST, CK, cTnT and expression of Bax protein(
P
<
0.05), as well as decreased protein expression of Shh,Gli1 and Bcl-2 (
P
<
0.05).Compared with the model group, the infarction area of rats was significantly reduced after treated with different doses of pachymoic acid, the myocardial fibers were arranged slightly neatly, the inflammatory cell infiltration was significantly reduced, with a decreased apoptosis rate, significantly low serum AST, CK, cTnT levels and Bax protein expression (
P
<
0.05), as well as significantly increased protein expression of Shh, Gli1 and Bcl-2 (
P
<
0.05).Compared with the high dose pachymic acid group, the myocardial infarction area of rats in the high-dose poria acid+cyclopamine group was significantly increased, myocardial fiber arrangement was disordered, inflammatory cell infiltration was significantly increased, with a increased apoptosis rate, significantly high serum AST, CK, cTnT levels and Bax protein expression (
P
<
0.05), as well as significantly decreased protein expression of Shh, Gli1 and Bcl-2 (
P
<
0.05).
Conclusion
2
The pachymoic acid can improve the myocardial ischemia-reperfusion injury in rats, which may be achieved by activating Shh/Gli1 signaling pathway.
