Effect of human cytokine-induced killer(CIK)cells on the model of human hepatocellular carcinoma in nude mice(LCI-D20)
Objective:To investigate the cure effect of human cytokine-induced killer (CIK) cells on the model of human hepatocellular carcinoma in nude mice. Methods:Peripheral blood mononuclear cells(PBMC)were obtained from healthy volun- teers by Ficoll gradient centrifugation and CIK cells were induced by culturing PBMC in the completed medium containing rhIFN-γ
rhIL-2 and mAb against CD3.By intraperitoneal injection of CIK cells in LCI-D20
the effect of prevention and therapy to HCC in LCI-D20 were assessed. Results:The weight of tumor in the CIK cells group and negative group was (1.125±0.363)gram and(2.682±0.526)gram respectively(t=6.06
P<0.01). The amount of AFP in blood of the group of CIK cells was(48.6±5.2)μg/L as the amount in the negative group was (82.2±7.2)μg/L(t=22.07
P<0.01). The metastasis rate in liver was 16.7% and 58.3% between two groups(χ2=4.44
P<0.01). The survival time of model in the therapy group were(73.3±7.03) days in the CIK group as compared with (52.3±5.24)days in negative group
(t=17.54
P<0.01). Conclusion:CIK cells may prevent metastasis in the model of LCI-D20 and prolong the survival time.
