Objective
2
To investigate the repair effects of nanomaterial fullerenol on the liver and kidney impairments and hematological abnormalities in lead-poisoned rats and explore its possible mechanism.
Methods
2
Newborn Wistar rats (5±1 g
regardless of sex) were randomly divided into normal control group (
n
=10
normal feeding)
lead-poisoning model group (
n
=10
0.2% of lead acetate in drinking water) and fullerenol intervention group (
n
=10
0.2% of lead acetate in drinking water
intraperitoneal injection of 5 mg/kg fullerenol once every day for 10 d from the age of 3 weeks). When the rats were at 5 weeks old
they were sacrificed
and HE staining was used to observe the pathological changes in the liver and kidney tissues. And the hematological indexes were analyzed using routine blood test. Total nitric oxide (NO) concentration detection kit and Western blotting were employed to measure the concentration of NO and the expression of inducible nitric oxide synthase (iNOS)
respectively.
Results
2
Fullerenol greatly changed the morphological abnormalities of liver and kidney
including hepatocyte swelling
nuclear conde
nsation and glomerular atrophy. Meanwhile
fullerenol significantly increased the hematological indexes associated with anemia (all
P
<
0.05)
such as red blood cell (RBC)
hemoglobin (HGB)
mean corpuscle volume (MCV) and mean corpuscle concentration (MCHC). What's more
compared with the lead-poisoning model group (0.58±0.06)
the expression of iNOS in the brain tissue was significantly reduced in the fullerenol intervention group (0.39±0.05
P
<
0.05).
Conclusion
2
By regulating NO-NOS system
fullerenol repairs the damages caused by lead poisoning
including the morphological abnormalities of the liver and kidneys as well as the important hematological indexes related to anemia.
