Objective
2
To investigate the expression of B cell translocation gene 2 (BTG2) in the patients with hepatocellular carcinoma (HCC)
and its relationship with postoperative prognosis.
Methods
2
One hundred and twenty-five patients with hepatocellular carcinoma confirmed pathologically and undergoing surgical treatment in our hospital from January 2012 to December 2014 were enrolled in this study. Among them
101 cases had complete follow-up data. According to the protein expression of BTG2 evaluated by immunohistochemical assay
the subjects were categorized to 3 groups
that is
high (+ +/+ + +)
low (+) and negative expression (-) groups. The correlation of BTG2 expression level with clinical pathological factors and overall survival were analyzed. Twelve pairs of tumor and para-carcinoma tissues were employed to detect the mRNA level of BTG2 by RT-PCR.
Results
2
In the obtained tumor tissues from 125 case
s
there 6
13 and 103 cases respectively with high
low and negative expression of BTG2
and the positive rates were 22
14
41 respectively for the corresponding para-carcinoma tissues. The expression was significantly different between the tumor and para-carcinoma tissues (
Chi
square=28.102
P
<
0.001). The RT-PCR results showed that there were 8/12 pairs of tumor and para-carcinoma tissues with BTG2 expression level lower in the tumor tissue than the para-carcinoma tissue. Occurrence of tumor thrombus was significantly associated with BTG2 expression (
Chi
square=8.305
P
=0.013)
and the low BTG2 expression group had the highest incidences of tumor thrombus. The median overall survival (mOS) was 36 months in the low BTG2 expression group (95%
CI
: 8.436~63.564)
while there was no mOS reached in the high expression and negative groups (Log-Rank
Chi
square=4.512
P
=0.105). However
the OS was significantly longer in the high than low expression groups (
Chi
square=4.266
P
=0.039). And there was no significant difference between the high and negative groups (
Chi
square=2.729
P
=0.099)
or between the low and negative groups (
Chi
square=1.400
P
=0.237). Stepwise Cox regression analysis showed that differentiation grade
hepatocirrhosis
tumor thrombus and metastasis were independent prognostic factors for OS in the HCC patients.
Conclusion
2
BTG2 protein is lowly expressed in the HCC tissues
and its high expression indicates better prognosis. It can be used as a potential prognostic factor for HCC.
