目的
2
P
<
0.01)。
结论
2
OECs/VitroGel™ 3D-RGD
Objective
2
To investigate the effects of transplantation of olfactory ensheathing cells (OECs)/VitroGel™ 3D-RGD hydrogel composite on repair at 12 weeks after spinal cord injury in New Zealand rabbits.
Methods
2
A total of 75 New Zealand rabbits were randomly divided into the blank control group, the model group, the hydrogel-only group, the OECs group, and the composite group, with 15 rabbits in each group. Except for the blank control group, rabbits in the model group, the hydrogel-only group, the OECs group, and the composite group all underwent establishment of a T
9
spinal cord injury model by weight-drop contusion and received local injections of PBS, hydrogel, OECs suspension, or OECs/hydrogel composite, respectively. Hindlimb motor function o
f the rabbits was assessed by the modified Tarlov score at 1, 2, 4, 8, and 12 weeks after surgery. Hematoxylin-eosin staining was used to measure the percentage of spinal cord cavity area. Neurofilament-200(NF-200)/glial fibrillary acidic protein(GFAP) was used to assess axonal regeneration and glial scar formation, and cluster of differentiation 68(CD68)/interleukin-10(IL-10) immunofluorescence was used to detect inflammatory cell infiltration and anti-inflammatory factor expression.
Results
2
At 12 weeks after surgery, the composite group had higher Tarlov scores and lower percentage of spinal cord cavity area compared with the model group, the hydrogel-only group, and the OECs group (
P
<
0.01). The composite group had more NF-200-positive axons and less GFAP-positive area in spinal cord tissue compared with the hydrogel-only group and the OECs group(
P
<
0.01).The composite group showed lower CD68-positive cell density and higher mean optical density of IL-10 in spinal cord tissue compared with the hydrogel-only group and the OECs group(
P
<
0.01).
Conclusion
2
OECs/VitroGel™ 3D-RGD composite transplantation is more effective in promoting tissue repair, axonal regeneration, and motor function recovery after spinal cord injury in rabbits than transplantation of cells or hydrogel alone. These effects may be related to three-dimensional scaffold support, neurotrophic factor secretion, and improvement of the inflammatory microenvironment.
