Objective To search for optimal culture condition of influenza virus in the tender cells. Methods Influenza A (H3N2
H1N1) and B virus were inoculated in MDCK cells
MEK cells and FRhK-4 cells under different culture conditions to observe cytopathic effect (CPE)
hemagglutination titer. Results after incubated at 33 ℃ for 72 h
cytopathic effect of viral infection in all three kinds of cells was observed
and hemagglutination assay showed MDCK cells and FRhK-4 cells were more sensitive than MEK cells to influenza virus. It is the best culture condition that the cells aged 48 h in maintenance media containing 1.4‰ NaHCO
3
1.0 μg/ml trypsin-TPCK. Conclusion The optimal culture conditions for influenza virus in cells were showed in this study.
