Objective To investigate the characteristics and performance of demineralized bone matrix (DBM) combined with nanoscale self-assembling peptide RADA16-Ⅰ (RADA16-Ⅰ/DBM)
and to evaluate its application as an optimal scaffold for enriching nucleated cells in bone marrow in selective cell retention (SCR). Methods DBM was combined with RADA16-Ⅰ by a cation-triggered process. The pore sizes of RADA16-Ⅰ/DBM were measured using a 3D video microscope. The combination of RADA16-Ⅰ and DBM was examined by X-
ray photoelectron spectroscopy (XPS). The performances of RADA16-Ⅰ/DBM and DBM in enriching nucleated cells in bone marrow were tested by SCR. The structures of RADA16-Ⅰ/DBM and DBM that had enriched nucleated cells were observed under a microscope after HE staining. Results RADA16-Ⅰ/DBM had significantly smaller pore sizes than DMB (220.55±158.54 μm
vs
414.32±175.00 μm
P
<
0.05). RADA16-Ⅰ hydrogel could firmly adhere to DBM. RADA16-Ⅰ/DBM had a significantly higher enrichment ratio than DMB for nucleated cells in bone marrow (6.05±0.96
vs
2.27±0.41
P
<
0.05). Conclusion RADA16-Ⅰ/DBM possesses improved performance in enriching nucleated cells in bone marrow
and can be used as an effective bone-repair material.
