Objective To observe the dynamic changes of proinflammatory cytokines IL-1β
IL-6 and anti-inflammatory cytokines IL-10 in LPS-induced RAW 264.7 cells and observe the effects of cholecystokinin o
ctapeptide (CCK-8) on their expression. Methods After RAW 264.7 cells were stimulated with 1 mg/L LPS for 0

mol/L respectively. Cytokines protein and mRNA expressions were assayed by ELISA and RT-PCR. Results The expression of IL-1β
IL-6 and IL-10 mRNA and protein were induced by LPS in RAW 264.7 cells in a time-dependent manner. CCK-8 inhibited LPS-induced IL-1β and IL-6 expression and augmented IL-10 expression in a dose-dependent manner. When the cells were treated for 24 h
CCK-8 augmentation resulted in higher LPS-induced IL-10 expression when compared with that after 48 hours’ treatment. Conclusion CCK-8 may participate in the anti-inflammatory response by inhibiting the expression of proinflammatory cytokines IL-1β and IL-6
meanwhile augmenting the expression of anti-inflammatory cytokine IL-10 induced by LPS.
