摘要

目的探索脂多糖（lipolysaccharide，LPS）干预气道上皮细胞细胞因子的分泌及蛋白激酶C(protein kinase C， PKC)的表达。方法培养A549细胞株，0、0.1、1.0、10.0、20.0、30.0、50.0、100.0 μg/mL的LPS干预A549细胞2、4、8、24、30、48 h以后收集细胞上清液及细胞质总蛋白，ELASA法检测A549细胞IL-1、IL-6、IL-8、IL-11的分泌，Western blot法检测相应的PKC的表达。结果不同浓度LPS干预A549细胞相同时间后细胞因子IL-1、IL-6、IL-8、IL-11的分泌量及PKC的表达量之间的差异均有统计学意义（

0.01）；PKC的表达和IL-11的分泌之间具有相关性（

0.05）。结论 LPS干预细胞因子IL-1、IL-6、IL-8、IL-11的分泌及PKC的表达，且LPS干预细胞因子IL-11的分泌可能和PKC信号通路有关。

Abstract

Objective To explore the expression of protein kinase C (PKC) and cytokines in airway epithelial cells interfered by lipopolysaccharide (LPS). Methods A549 cell line was cultivated and incubated with different concentrations of LPS for 2

30 and 48 h

respectively. The culture medium

and whole cell proteins were extracted

and the levels of cytokines including interleukin (IL)-1

IL-6

IL-8 and IL-11 secreted by A549 cells were detected by enzyme-linked immunosorbent assay (ELISA). Meanwhile

the expression of PKC was detected by Western blotting.

Results There were significant differences in the levels of IL-1

IL-6

IL-8

IL-11 and expression of PKC in the A549 cells interfered by various concentrations of LPS at the same time point (all

values

0.001). The expression of PKC was correlated with the level of IL-1 (

0.05). Conclusion The secretion of IL-1

IL-6

IL-8 and IL-11 and expression of PKC are interfered by LPS. The secretion of IL-1 interfered by PKC may be correlated with PKC signal pathway.
