Objective To prepare a new type of lipid-coated ultrasound microbubble loaded with oxygen
and to study its basic characteristics. Methods A new type of ultrasound microbubble carrying oxygen was prepared by mechanic vibration. The appearance
concentration
mean diameter and zeta potential of the oxygen-loaded microbubbles were investigated
and their changes within 7 d and after
60
Co ray irradiation were observed. The microbubbles were mixed with oxygen-deprived solution and the dissolved oxygen (DO) concentration was compared before and after
ultrasound irradiation (5 kHz
3.5 W). The enhancing effect of ultrasound imaging on rabbit liver was evaluated after the microbubble injection. Results The oxygen-loaded microbubbles appeared as a uniform size with a mean diameter of 959.7±161.2 nm
concentration of (4.01±0.69) ×10
9
/mL and zeta potential of -(12.9±2.4)mV. Their appearance
concentration
mean diameter and zeta potential didn′t change within 7 d or after radiosterilization of
60
Co ray (
P
>0.05). The DO concentration of the solution significantly increased after ultrasound irradiation. The ultrasound imaging on rabbit liver was greatly enhanced after oxygen-loaded microbubble injection. Conclusion The new type of self-made lipid-coated oxygen-loaded microbubble has stable performance and good capacity of carrying and releasing oxygen
and it can be used as a novel and ideal ultrasound contrast agent.
