Objective To investigate the role of cancer-associated fibroblasts (CAF) in non-small cell lung cancer (NSCLC) and possible mechanisms. Methods Primary cultures of CAF and normal fibroblasts (NF) from NSCLC patients were established. The effect of CAF on the growth of NSCLC was observed. The expression of interleukin (IL)-6 and related molecules were detected by ELISA. The effect of IL-6 on the proliferation of NSCLC cells was evaluated by MTT assay. Results CAF had greater ability than NF to enhance the tumorigenicity of lung cancer PC9 cell lines. IL-6 was highly expressed in the CAF conditioned media
and further studies identified that IL-6 could promote the growth of NSCLC. CAF also can promote xenograft tumor growth in a mouse model. Conclusion CAF can promote the growth of NSCLC by secretion of IL-6.
