Objective To determine the effects of myricetin on oxidative stress in vascular endothelial cells induced by high glucose. Methods Human umbilical vascular endothelial cells (HUVECs) were treated by 5.56 mmol/L glucose as normal control
5.56 mmol/L glucose plus 27.8 mmol/L mannitol as an osmotic control
or 33.36 mmol/L glucose for 48 h in the absence and presence of myricetin (0.1 to 10 μmol/L). Cell viability and apoptosis
were detected by CCK-8 assays and annexin V-FITC/PI double staining using flow cytometry
respectively. In addition
the intracellular reactive oxygen species (ROS)
malondialdehyde (MDA) content and the activities of superoxide dismutase (SOD) and catalase (CAT) in the culture supernatants were measured. Results Compared with the normal control group
high glucose significantly decreased cell viability and the activities of SOD and CAT (
P
<
0.05)
while increased the apoptotic rate
intracellular ROS and MDA contents in HUVECs (
P
<
0.05). However
myricetin pretreatment significantly inhibited these high glucose-induced above phenomena (
P
<
0.05). Conclusion Myricetin attenuates high glucose-induced oxidative stress and apoptosis in vascular endothelial cells.
