Objective      To determine the effect of the regulation of Rab26 expression on the apoptosis and migration of human HeLa cells.  Methods      Eukaryotic expression vectors containing constructs of Rab26 cDNA or Rab26 siRNA were transfected into HeLa cells for overexpression or silencing of Rab26

respectively. Untreated HeLa cells and cells transfected with empty vectors were used as normal control and negative control. At 48 h after transfection

the protein  and mRNA levels of Rab26 were determined by Western blotting and RT-PCR analysis. Cell apoptosis was assayed by flow cytometry and migration rate was examined by scratch wound assay. Results     Compared with the negative control and normal control

Rab26 expression was significantly increased in the Rab26 cDNA-overexpressed group but decreased in the Rab26 siRNAmediated group (P0.05). Also

Rab26 overexpression significantly enhanced cell apoptosis and inhibited migration rate(P0.01)

whereas siRNA-mediated downregualtion of Rab26 exerted the adversely effects (P0.05).  Conclusion     Rab26 expression affects the apoptosis and migration of HeLa cells

which supports the notion that regulation of Rab26 expression may be a novel target for treatment of cervical carcinoma.
