Objective To screen the differentially expressed genes related to acting transcription factor 3 (ATF3) in the lung tissue of mouse model of lipopolysaccharides (LPS) induced acute lung injury (ALI) with GeneChip assay. Methods RNAs were isolated from lung tissues of wild-type (WT) mice and ATF3 knockout (KO) mice after LPS treatment. Differentially expressed genes were screened out by GeneChip array. Key genes were selected for further validation by real time (RT)-PCR. Results Analysis of genomic screen data showed that a total of 1 692 genes were identified differentially expressed in ATF3 KO mice compared with WT mice. Among them
21 genes were significantly-related to inflammatory regulation
including 18 up-regulated genes Myo18a

and 2 down-regulated genes Padi4 and Wfdc8.The up-regulation of Tnfsf15 (also named TL1A) was verified by RT-PCR showing elevated mRNA expression. Conclusion TL1A maybe involved in the ATF3 mediated antiinflammation of LPS-induced ALI.
