Objective To investigate the inhibitory effect of Jumonji domain-containing protein 2B (JMJD2B) siRNA on biological behaviors in human squamous carcinoma cells. Methods The expression of JMJD2B in the human squamous cell carcinoma tissue and A431 cells was detected by immunohistochemical staining and cell immunofluorescence assay. The A431 cells were transfected with JMJD2B siRNA by Liposome. Then the proliferation
colony formation
cell apoptosis
cell cycle
migration and invasion were measured by CCK-8 assay
colony formation assay
flow cytometry
and transwell migration assay
respectively.
Results JMJD2B was highly expressed in the tumor tissues than normal skin tissues. Compared with the un-transfected A431 cells
the cell proliferation
colony formation
and migration and invasion abilities were significantly decreased
while cell apoptosis was increased in the cells after siRNA transfection (P0.05). Cell cytometry indicated JMJD2B siRNA transfection arrested the cells at G
1
phase (P0.05). Conclusion JMJD2B is highly expressed in the human squamous cell carcinoma. JMJD2B siRNA promotes cell apoptosis
and suppresses cell proliferation
colony formation
and migration and invasion abilities in A431 cells.
