Objective To detect the brain expression levels of ferroportin 1 (Fpn1) and ceruloplasmin (CP) in rats at early period after experimental subarachnoid hemorrhage (SAH) and discuss the mechanisms of iron metabolism disorder in early brain injury (EBI). Methods A total of 96 male adult Sprague-Dawley rats were randomly divided into 2 groups: the SAH group and sham operation group. The SAH rat model was established by injecting fresh arterial
non-heparinized blood into the prechiasmatic cistern to induce subarachnoid hemorrhage. Thirty-six animals were sacrificed in 24 h after SAH or sham operation
and the left 12 were killed in 24 for brain water content. Western blotting and immunohistochemical assay were used to examine the expression of Fpn1 and CP. Evans Blue perfusion was used to detect the permeability of blood-brain barrier (BBB). Wet and dry weight method was used to determine the ratio of brain water content in 72 h after SAH. Results Compared with the sham group
the brain water content of the rat brain tissue and the BBB permeability were significantly increased (P0.05). Western blotting and immunohistochemical assay showed that the expression levels of Fpn1 and CP in the cerebral cortex and hippocampus were significantly down-regulated in the SAH group than the sham group (P0.05). Conclusion The expression of iron metabolism-related proteins Fpn1 and CP is obviously inhibited in SAH rat model.
