Objective To investigate the anti-hepatofibrotic effects of geniposide on liver fibrosis. Methods Male C57 mice were induced with 20% CCl
4
(2 mL/kg) for 6 weeks to induce liver fibrosis
and were treated with intragastrical gavage of 50 mg/kg geniposide for 14 d since the fifth week of CCl

induction. Liver fibrosis of the mice was evaluated by HE staining and Sirius Red staining. Activities of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST)
and liver content of hydroxyproline were measured with automated biochemistry analyzer. The protein levels of transforming growth factor beta1 (TGF-β1) and α-smooth muscle actin (α-SMA) were detected by Western blot analysi
s. Results Geniposide significantly attenuates the progression of liver fibrosis induced by CCl
4
. Treatment with geniposide also resulted in significant decreases in the serum levels of ALT and AST
as well as the content of hydroxyproline after CCl
4
induction(
P
0.01). Geniposide significantly reduced the protein expression levels of TGF-β1 and α-SMA in the liver(P0.01). Conclusion Geniposide exerts anti-fibrotic effects on the liver induced by CCl
