Objective To observe the effects and possible mechanism of Ln5γ2 on the invasion and migration in SGC-7901 cells. Methods Four groups of gastric cancer SGC-7901 cells were studied
including an Ln5γ2 chemotaxis group
an EGFR interference group
an empty vector group and a control group. After Ln5γ2 chemotaxis of SGC-7901 cells
cell numbers passing through the Transwell membrane with or without Matrigel glue were observed. The concentrations of MMP-2 and MMP-9 in the cell buffer were tested by ELISA. The cell skeleton changes of the four groups were examined by laser co-focal microscopy. Results The numbers of cell passing through the Transwell membrane with or without Matrigel glue in the Ln5γ2 chemotaxis group were greater than those of other groups (P0.01). However
the numbers showed no difference after EGFR interference (P0.05). MMP-2 and MMP-9 contents in the EGFR interference group declined obviously. The structure of cell skeleton was faintness and the formation of pseudopodium was reduced. Conclusion Ln5γ2 improves the invasion and migration of SGC-7901 cells. The possible mechanism is Ln5γ2 binding to EGFR
then changing the cell skeleton and reducing MMPs secretion.
