Objective To observe the expression of L amino acid transporter 1 (LAT1) and its heterodimer CD98hc in gastric cancer SGC-7901 cells and the effect of LAT1 on proliferation and migration of SGC-7901 cells. Methods Gastric cancer SGC-7901 cells were divided into 1 μmol/L BCH intervention group,10 mol/L BCH intervention group,100 μmol/L BCH intervention group and control group. Expression of LAT1 and CD98hc mRNA in gastric cancer SGC-790
1 cells was detected by RT-PCR
IF and Western blotting
respectively. After the SGC-7901 cells were cultured with BCH at different concentrations for 48 h
their proliferation was detected by MTS colorimetry
distribution of cell cycle in different groups was assayed by flow cytometry
and cell migration in different groups was observed in Transwell chamber. Results LAT1 and CD98hc were highly expressed in SGC-7901 cells. The proliferation and migration level in SGC-7901 cells decreased with the increasing BCH concentration and were significantly lower in 3 BCH intervention groups than in control group. Cell cycle analysis showed that the number of SGC-7901 cells was lesser in S phase and greater in G

phase after BCH treatment(P0.05). Conclusion LAT1 and CD98hc are expressed in SGC-7901 cells. LAT1 may promote the proliferation and migration of SGC-7901 cells and speed up their cell cycle process.
