Objective To study the correlation between synthesis diagnostic indications (SDI) of ultrasound-guided diffused optical tomography (DOT)
expression of hypoxia inducible factor-1α (HIF-1α)
and microvessel density (MVD) in breast cancer. Methods SDI were demonstrated by ultrasound-guided DOT for 131 patients with benign mass and 142 patients with breast cancer admitted to our department from November 2008 to September 2010. Optimal diagnostic threshold of breast cancer was established by receiver operating characteristic (ROC) curve. Expressions of HIF-1α and MVD in breast cancer were detected with the immunohistochemical method. Correlation of SDI with expression of HIF-1α and MVD in breast cancer
and pathological features of breast cancer was analyzed. Results The SDI value was significantly lower in patients with benign breast mass than in those with breast cancer (109.35±48.03 vs 190.87±60.33
P0.05). ROC curve analysis showed that the SDI value greater than 141.2 was used as the optimal diagnostic threshold of breast cancer. The SDI and MVD values were significantly higher in breast cancer with positive HIF-1α expression than in that with negative HIF-1α expression (P0.05). The SDI value was positively correlated with the MVD value (r=0.673
P0.01). The SDI value and the expression of HIF-1α were significantly correlated with the tumor size
lymphatic metastasis
TNM stage
and CerbB-2 expression (P0.05)
but not with the ER and PR expression. Conclusion As a new function testing technique
ultrasound-guided DOT plays an important role in diagnosis of breast cancer.
