Objective To investigate the inhibitory effect of cFLIP antisense oligonucleotide (cFLIP-asODN) on the proliferation of human colon carcinoma cell line SW620. Methods Human colon carcinoma cell line SW620 was transfected with cFLIP-asODN by liposome Lipofectamine
TM
2000. Besides
a cFLIP-asODN group
a liposome+nonsense ODN (NSODN) group and a blank control group were constructed as the control groups. Inverted fluorescence microscope was used to detect the transfection efficiency of the complexes. Real-time
quantitative RT-PCR and Western blotting were used to detect the expression level changes of cFLIP mRNA and protein in SW620 cell after transfection. MTT assay was used to analyze the effects of the complexes on the proliferation of SW620 cell. Flow cytometry was used to determine the apoptosis of SW620 cell after transfection. Results The proliferation of SW620 cell transfected with cFLIP-asODN delivered by liposome Lipofectamine
TM
2000 was significantly inhibited (P0.05)
as compared with the three control groups. The cFLIP mRNA and protein expression levels of transfected SW620 cell were significantly lower than those of non-transfected SW620 cell. The apoptosis of SW620 cell in the liposome+cFLIP-asODN group was significantly higher than those in the three control groups [(29.5±2.4)% vs (5.8±0.7)%
(5.5±0.8)% and (5.2±0.5)%; P0.05
]
. Conclusion In vitro liposome-mediated transfection with cFLIP-asODN can significantly inhibit the proliferation of human colon carcinoma cell line SW620.
