Objective To determine the effects of anti-survivin oligonucleotides (ASODN) on the proliferation
apoptosis
invasion and migration in gastric cancer BGC-823 cells. Methods After ASODN or SODN was introduced by Lipofectamine
TM
2000 transfection reagent into BGC-823 cells
the mRNA expression of survivin
was detected by RT-PCR
cell proliferation was measured by 4-methyl-tetrazolium (MTT) assay
cell
invasion and migration were assayed by Transwell chamber tests
and cell cycle and apoptosis index (AI) was detected by flow cytometry. Results ASODN transfection resulted in a marked decrease of survivin expression (P0.05). And the cell proliferation was inhibited by ASODN transfection in a dose- and time-dependent manner (both P0.05). Both cell invasion and migration activities of BGC-823 were decreased (P0.05) after the transfection of 600 nmol/L ASODN. AI of ASODN transfection group was increased significantly when compared with that of control group (11.31±0.38 vs 1.62±0.36
P0.01)
and proliferation Index (PI) was decreased significantly in ASODN transfection group than in control group (27.77±2.97 vs 43.80±0.81
P0.05). Cells in G
2
/M stage were increased in ASODN transfection group compared with control group (P0.01). Conclusion Down-regulation of survivin by ASODN inhibits the invasion and migration of BGC-823 cells through suppressing cell proliferation and improving cell apoptosis.
